The Long Tail of mRNA Regulation

چکیده

Long stretches of RNA trail after the coding region in most eukaryotic mRNAs. These 3' untranslated regions (3' UTRs) hold the key for many aspects of regulated gene expression, influencing mRNA stability, protein synthesis, and ultimately protein function. Lara Szewczak asked Rachel Green, Ian Macrae, and Christine Mayr about how dynamic regulation by proteins and microRNAs works within 3' UTRs. An annotated excerpt of the conversation appears below, and the full conversation is available with the article online.

Purification of Saccharomyces cerevisiae eIF4E/eIF4G/Pab1p Complex with Capped mRNA

Protein synthesis is one of the most complex cellular processes, involving numerous translation components that interact in multiple sequential steps. The most complex stage in protein synthesis is the initiation process. The basal set of factors required for translation initiation has been determined, and biochemical, genetic, and structural studies are now beginning to reveal details of their...

Regulated nuclear polyadenylation of Xenopus albumin pre-mRNA.

Cytoplasmic regulation of the length of poly(A) on mRNA is a well-characterized process involved in translational control during development. In contrast, there is no direct in vivo evidence for regulation of the length of poly(A) added during nuclear pre-mRNA processing in somatic cells. We previously reported that Xenopus serum albumin [Schoenberg et al. (1989) Mol. Endocrinol. 3, 805-815] an...

The Response of Skeletal Muscle-Expressed Neurotrophins to Acute Resistance Exercise in Male Wistar Rats

Background. BDNF and NT-4/5 have been proposed to be involved in the coordinated adaptations of the neuromuscular system to the elevated level of activity, but an activity-dependent expression of neurotrophins in skeletal muscle is not well established. Objectives. We, therefore, investigated the effect of one session of resistance exercise on mRNA expression of some neurotrophins in Slow and ...

RNA-Binding Proteins which Affect mRNA Translation or Stability

The regulation of gene expression can take place at many stages subsequent to transcriptional activation. In eukaryotic systems the messenger RNA (mRNA) is subject to processing, nucleocytoplasmic transport, translation and degradation. Each of these points serves as a checkpoint at which the ultimate production of encoded protein can be regulated. Studies in the past two decades have shown tha...

Hydroalcoholic Extract of Anchusa Italica Protects Global Cerebral Ischemia-Reperfusion Injury Via a Nitrergic Mechanism

Introduction: In stroke models, Inducible Nitric Oxide Synthase (iNOS) expression initiates cellular toxicity due to excessive Nitric Oxide (NO) generation. Anchusa italica is a medicinal herb with anti-inflammatory, antioxidant and neuroprotective properties. This study evaluated the antioxidant activity and NOS mRNA expression of the Hydroalcoholic Extract Of Anchusa Italica (HEAI) in an expe...

بررسی تاثیر یک جلسه فعالیت مقاومتی بر بیان ژن های NT-4/5 و p75 در عضلات سریع و آهسته موش های صحرایی

Background and purpose: Activity-dependent expression of neurotrophins in skeletal muscle is not well established. In this research we aimed at studying the effect of one session resistance exercise on mRNA expression of NT4.5 and P75 proteins in slow and fast skeletal muscles of Wistar rats. Materials and methods: Sixteen male Wistar rats (10 wk of age) were housed at room temperature under a...
